Radiolysis of DNA in the proteins nucleic-acids complexes

Abstract :

We report here a physico-chemical and a molecular study of the radiolysis of DNA interacting with regulatory proteins (lac repressor and CRP), histones (in the core nucleosome) or a bacterial chromosomal protein. Our results show a very efficient protection of the binding site on DNA by the bound proteins. The radioprotection results from physical screening against the attack by the radicals radioinduced in water. The results are independent of the quality of the radiation (beta-rays, -gamma-rays and fast neutrons). A new method of molecular footprinting using radioinduced radicals as chemical probes, and suitable for in vivo studies, is proposed.
